I have been off loestrin generic for like three months and I'm getting back on it next month, and I would like to know how soon does it become effective?

It will be effective quite soon however it is not at the full effectiveness until your body begins to adjust to it. The absolute recommended time is to keep using a condom for at least a week when you start the pill, however some doctors will advise people to try for 2 weeks with a condom just for that added protection.

So I should wait at least a week, but two weeks is safe? So the effect of the medicine kicks in after two weeks?

It kicks in within the first day, however to put your mind at ease of any worry I would use a condom as added protection for at minimum a week. If you decide to use one longer for again ease of mind then that is okay as well. However the entire effectiveness is based on you taking it routinely. Missing, skipping, forgetting, or taking the pill at random times will all throw the effectiveness off. Have a time set out to take it, and try to get as close to that time every single day. There is about a 3 hour window but it is necessary to be taking it around that same time everyday.
